Growth parameters of Dendrobrium parishii protocorms in transflasking media modified with coconut milk and banana juice


Abstract
Protocorms culture of Dendrobium parishii (Dendrobium parishii Rchb.f.) grown on Vacin and Went-based transferring media modified by the mixture of coconut milk and banana juice were studied. The objectives of this study were to find transferring media suitable for the culture. Four treatments consisting of Vacin and Went, Vacin and Went plus coconut milk, plus banana juice and coconut milk were laid out in a completely randomized block design (CRD) with four replications. Sprouts of the Dendrobium parishii seeds were allowed to grow in the Vacin and Went for 2 months and then transferred into the treatments for 6 months. Data were recorded for the stem-high and weight, the number of roots and the number of leaves of protocorms in vitro.
The results of protocorms of Dendrobium parishii were best growing
in the mixture by coconut milk, banana juice on media transfer Vacin and Went. Especially the stem-high average, the weight average, the number of roots average and the number of leaves average were advantage.


Introduction

Dendrobium parishii is the most popular a wild orchids in Thailand. This genus has beautiful flowers, which is one important orchid grown for commercial production of potted plants. Dendrobium parishii is a compact epiphyte from the northern and north-eastern Thailand. It grows in deciduous forest at moderate altitudes (250-1500 m), where the climate is strongly seasonal with a dry period in late winter. The flowering season is middle summer.
We can produce and raising the Dendrobium parishii from their seeds in capsule on nutrient media (Vacin and Went). The germination was successful to be protocorms, after about 2 months the seeding should be large enough to be removed from flask and they should have sent to a new nutrient media. As soon as the protocorms were strong enough and were growing well-probably after about 6 months. We removed them from the propagator to outside area in cultivation. But the majority of protocorms in the new media were die after 3 months.
Coconut milk and Banana powder were used in culture media for orchid seeding. Some growers homogenized banana fruit pulp with their media, whereas others stirred puree into their solutions. Opinion varied at the time as to whether green bananas enhanced growth better than the ripe ones, and preferences still exist among growers. Over the years orchidologists have devised a number of nutrient formulae for the aseptic germination of a large variety of orchid genera. Coconut milk in the formulae contain was the one ingredients. It was highly effective on variety genera.
Thus, the objectives of experiment were to study the growing of Dendrobrium parishii protocorms in coconut milk, banana juice on media transfer Vacin and Went.

Methodology

1. Plant material

Protocorms were induced from seed of Dendrobrium parishii .It was raising on Vacin and Went-based for 2 months.

2. Media transfer.
Four treaments medias were :
a) Nutrient media Vacin &Went the pH was between 4.8-5.2
b) Coconut milk 150 ml/L + Vacin &Went
c) Banana juice 150 ml/L + Vacin &Went
d) Coconut milk : Banana juice = 75 :75 ml/L + Vacin &Went.

3. experimental
Seeds in vitro germinated in nutrient media Vacin &Went for 2 months. The 40 completely protocorms was selected. The experiment was designed in CRD with four replications with media transflasked. And stem-high and weight, the number of roots, the number of leaves of protocorms in vitro were recorded
Result , discussion and conclusion:
Seeding sub cultured in only Vacin & Went media and modified Vacin & Went media added with banana juice 150 ml/L, coconut milk150 ml/L and mixed blended banana with coconut milk 75:75 ml/L for 6 months. Table 1, 2, 3 and 4 showed the four treatments Medias were:


The results showed average plant height and weight, the number of roots and leaves on Vacin & Went media without modification as lowest. However, the media Vacin & Went media modification with banana and coconut milk was showed the best result.

รูปข้างบนเป็นกล้วยไม้สกุลหวาย เป็นไม้ในฟาร์ม

กล้วยที่ผมทำ เป็นกล้วยไม้ป่า มีชื่อเรียกกันทั่วไปว่า เอื้องสายน้ำครั่ง พบมากบริเวณทางภาคเหนือ และภาคตะวันออกเฉียงเหนือตอนบน ในธรรมชาติมีการกระจายพันธ์ด้วย เมล็ด โดยฝักแก่แตกออก แล้วกระจายไปตามลม ซึ่งมีโอกาสเป็นต้นอ่อนได้ยากมากเนื่องจากข้อบกพร่องจากเมล็ดของมันและสภาพแวดล้อมที่เปลี่ยนแปลงไป
ด้วยความที่มีดอกสวยงาม จึงมีการลักลอบออกจากป่าเป็นจำนวนมาก ทำให้กลายเป็นกล้วยไม้ที่เกือบสูญพันธ์ เช่นเดียวกับรองเท้านารีเหลืองเมืองกาญจน รองเท้านารีดอยตุง และอีกหลายๆ สายพันธุ์ เป็นเรื่องน่าเสียดาย หากพืชเหล่านี้หายไปจากเมืองไทยจริงๆ

ความเห็นส่งไปทางเมล์แล้ว บางส่วนตอบในนี้เพราะเห็นว่าอาจเป็นประโยชน์กับคนอื่น ๆ ด้วย ก่อนอื่นอยากถามว่าเวลาที่ต้องส่ง paper มีอีกสักเท่าไหร่ เพื่อจะพัฒนาให้สัมพันธ์กับเวลาที่มี ผมคงช่วยในส่วนพัฒนาเปเปอร์ และขอให้คุณหมอดูภาษาอังกฤษให้น่าจะดีกว่าครับ

1. ส่วนบทคัดย่อ คงค้างไว้ก่อนนะครับ เพราะข้อมูลยังไม่ลงตัว แล้วค่อยกลับมาดูใหม่

2. ส่วน คำนำ การบ้านคุณทำน้อยไปนิดนึง คิดว่าน่าจะปรับปรุงได้ เขียนบอกความสำคัญของเรื่องที่ทำ บอกสถานภาพของการวิจัย โดยมีการอ้างอิงเอกสารที่สมบูรณ์ บอกประเด็นคำถามและสมมุติฐาน และวัตถุประสงค์ สุดท้ายบอกว่าผลการศึกษานี้มันมีประโยชน์อะไร

3. ส่วนอุปกรณ์และวิธีการ เขียนหัวข้อย่อยมากไปแต่ก็ยังไม่ชัดเจน ตรงนี้เป็นส่วนที่จะให้ความรู้กับผู้อ่านได้ด้วย ทำให้เปเปอร์มีคุณค่ามากขึ้น โดยหลักแล้วคุณทำมาอย่างไรก็อธิบายให้ชัดเจนให้คนอ่านนึกภาพออกและทำตามได้

คุณคัดเลือกแหล่งของเมล็ดอย่างไร ฝักที่แตกแล้ว ฝักสด คุณผสมพันธุ์เองหรือเปล่า อายุกี่วัน หรือเก็บมาจากป่า จากแหล่งไหน จังหวัดไหน

คุณ เพาะเมล็ดอย่างไร ทำความสะอาดผิว การทรีตสารเคมี และอื่น ๆ เมล็ดมีความงอกเป็นอย่างไร และมีการพัฒนาอย่างไร กี่ระยะ

คุณคัดเลือกหน่วยทดลองอย่างไร สม่ำเสมอแค่ไหน ขนาดหน่วยทดลองมากหรือน้อย (กี่ต้นต่อหน่วย)

สภาพการทดลองเป็นอย่างไร ควบคุม แสง อุณหภูมิ ความชื้นอย่างไร ความยาวของกลางวันกลางคืนเป็นเท่าใด

คุณวัดแต่ละพารามิเตอร์อย่างไร

และสถิติที่ใช้ ในกรณีนี้ คุณทำ combined analysis of variance ได้ จาก 3 ครั้งที่คุณประเมิน ถ้า interactions ระหว่าง treatment และ เวลาประเมิน ไม่มีนัยสำคัญ และ error variance มี homogeniety และคุณเปรียบเทียบค่าเฉลี่ยอย่างไร

4. ผลการทดลอง คุณอาจเสนอตั้งแต่ การเพาะเมล็ด การเปรียบเทียบสูตรอาหาร จนกระทั่งการอนุบาลในเรือนทดลอง ผมเข้าใจว่าคุณทำมาทั้งหมดนั่นแหละ

ดูข้อมูลทั้งหมดก่อนแล้วค่อยพิจารณาว่าจะเสนออะไร แบบไหน จะเดินเรื่องอย่างไร อาจต้องทำคอมไบน์ อะนาไลซิส ถ้าทำได้ อธิบายผลได้ง่ายขึ้น ประหยัดพื้นที่ตารางลงมาก ในขั้นแรกมี 4 ตาราง ผมลองเปลี่ยนรูปตารางให้แล้ว อาจเปลี่ยนเป็นเสนอแค่ 3 ตาราง ตามเวลาที่ประเมิน แทนที่จะแยกตามพารามิเตอร์

อื่น ๆ ผมอธิบายในเท๊กซ์ไปแล้ว การอภิปรายผล ต้องรอดูการเขียนผลให้ลงตัวก่อนครับ

ตามที่คุณ xx บอก ผมเข้าใจดังนี้ครับ คุณจะเสนอเป็นโปสเตอร์ในภาค poster และ abstract ในภาค oral presentation ซึ่ง abstract นั้น proceedings อาจจัดพิมพ์ทีหลัง ส่วน paper จะเป็น handout เสริมในภาค poster และจะไม่มีการตีพิมพ์

ผมดูข้อมูลแล้ว ถ้ามีการเพิ่มเติมข้อมูลบ้างจากส่วนการเพาะเมล็ด และ การอนุบาล คุณสามารถพัฒนาเป็น paper ที่ดีขึ้นได้ และตีพิมพ์ในวารสารดี ๆ หน่อย วารสารที่เหมาะสมผมเสนอไว้ 3 ชื่อคือ Songklanakarin Journal of Science and Technology, Asian Journal of Plant Science ( ของไทย)และ Pakistan Journal of Plant Science ซึ่งผมคิดว่าเหมาะสมกับเนื้อหาและไม่ต่ำเกินไปนัก เพราะคุณไม่ได้ส่ง proceedings เป็น paper เต็ม ดังนั้น paper นี้ ลงวารสารได้ ถ้าคุณสนใจจะทำ

ประโยชน์ของ paper นี้ คือช่วยลดเวลาสำหรับการขยายพันธุ์กล้วยไม้หายากเพื่อการอนุรักษ์เชื้อพันธุกรรมในสภาพนอกธรรมชาติและเป็นการวิจัยพื้นฐานเพื่อการปรับปรุงพันธุ์ คุณต้องชูประเด็นนี้เป็นจุดขาย

คุณอาจเสนอแบบ 3 ตาราง ตามเวลาที่คุณประเมินแทนที่จะเสนอตามพารามิเตอร์ คุณเลือกได้ว่าจะเป็น 4 หรือ 3

Introduction

Dendrobium parishii is the most popular wild orchid in Thailand. This genus has beautiful flowers, important in the commercial production of potted plants. Dendrobium parishii is a compact epiphyte from Northern and Northeastern Thailand. We are able to produce and raise Dendrobium parishii from seed in capsules on nutrient media of Vacin and Went. Germination to protocorms is usually successful. After about 2 months, the seeding are usually large enough to be removed from the flasks and transferred to a new nutrient media. As soon as the protocorms are strong enough and growing well, usually after about 6 months, they are moved from the propagator to an outside area in cultivation. The problem is that the majority of protocorms in the new media will die after 3 months.

Coconut milk and banana powder have previously been used in culture media for orchid seeding. Some growers homogenized banana pulp with their media, whereas others stirred banana puree into their solutions. Opinion difffer as to whether green bananas or ripe banana were better growth enhancers, and preferences still exist among growers. Over the years, orchidologists have devised a number of nutrient formulae for the aseptic germination of a large variety of orchid genera. Coconut milk was the one ingredients that was highly effective on a variety of genera of orchids.

The objective of this experiment was to study the effects of coconut milk and banana juice as an additive to Vacin & Went transfer media on the growth of Dendrobrium parishii protocorms.

Methodology

1. Plant material

Protocorms were induced from seeds of Dendrobrium parishii grown on Vacin and Went-based media for 2 months.

2. Transfer media.
Four kinds of transfer medias were used:
a) Standard Vacin & Went media with pH between 4.8-5.2
b) Coconut milk additive to Vacin & Went media, 150 ml/L
c) Banana juice additive to Vacin & Went, 150 ml/L
d) Coconut milk/banana juice additive to Vacin & Went media, 75/75 ml/L

3. Experiment
In vitro germination and growth of Dendrobrium parishiiin in Vacin & Went media was done for 2 months. Forty complete protocorms were selected. Completely randomized block design with four replications in transflasked media was done. Stem-height, weight, number of roots and number of leaves of the protocorms was recorded in vitro.

Results , discussion and conclusion:
Results of seeding grown in Vacin & Went as well as modified Vacine & Went mediae are shown in Tables 1, 2, 3 and 4.

Orchids grown on unmodified Vacin & Went media showed the lowest averages for plant height, weight, number of roots and leaves. Vacin & Went media modified with banana juice and coconut milk resulted in the best results.

ใจผมคิดว่าควรจะบอกด้วยว่า กะทิ กับ น้ำกล้วย ได้มาจากไหน กระป๋อง หรือ สด กล้วยอะไร กะทิเข้มข้นแค่ไหน อาจรวมกระบวนการผลิตกะทิหรือน้ำกล้วยด้วย (Coconut milk = กะทิ ส่วน Coconut water = น้ำมะพร้าว ฝรั่งมักงง ไม่รู้ความแตกต่าง)

ผมอ่านในเอกสารเกี่ยวกับการเพาะเมล็ดกล้วยไม้ มีการกล่าวถึง คำสามคำ คือ coconut milk และ coconut cream และ coconut water ผมยังเข้าใจว่า coconut milk คือน้ำมะพร้าว
coconut cream= น้ำกะทิ coconut water= เครื่องดื่มที่ทำจากน้ำมะพร้าว
วานท่านอาจารย์ทั้งสอง และผู้รู้ช่วยอธิบายความหมายของคำทั้งสามด้วยว่า แท้จริงแล้วมันคืออะไร

เท่าที่ผมใช้ coconut cream = หัวกะทิ คือคั้นครั้งแรกๆ ข้นหน่อย เปรียบเทียบกับ cream ที่ได้จากนม
ส่วน coconut milk = กะทิธรรมดา เปรียบเทียบกับนมสดธรรมดา
แล้ว coconut water = น้ำมะพร้าวที่เราทิ้ง นอกจากจะได้จากมะพร้าวอ่อน
coconut drink = อันนี้ไม่ค่อยแน่นอน แล้วแต่คนใช้ อาจเป็นน้ำมะพร้าวอ่อน มะพร้าวเผา หรือเครื่องดื่มที่ผสมกะทิก็ได้
